Isolation of a divergent gamma-delta T-cell receptor gene from mice.
A murine T-cell hybridoma was derived from the peripheral lymph nodes of BALB/c mice immunized with the tyrosinase peptide p1B.2. The T-cell hybridoma was selected for its ability to proliferate in response to tyrosinase peptide p1B.2. The hybridoma secreted gamma-interferon upon stimulation with p1B.2. The gamma-interferon secreted by the hybridoma was immunoreactive with monoclonal antibodies specific for gamma-interferon, suggesting that this hybridoma expressed gamma-interferon genes. Two gamma-interferon cDNA clones were isolated from the hybridoma library by screening with a synthetic peptide p1B.2 in the presence of a subtracted T-cell cDNA library. The cDNA contained a 5′ terminal oligo-d(T)20 sequence and an open reading frame encoding a polypeptide of 116 amino acids with a predicted molecular mass of 13.2 kDa. The predicted primary structure contained 5 characteristic motifs of the mammalian gamma-interferon gene, including the highly conserved 17-amino-acid cysteine-rich motif. The gene could be aligned with the human and murine genes for human gamma-interferon. Using two anti-murine gamma-interferon antibodies, it was found that the murine gamma-interferon cDNA-encoded protein could not be recognized by one of the antibodies, suggesting that the antigenic determinant of the protein could not be present on the corresponding peptide. We cloned a genomic DNA fragment containing the entire coding region of the gene. The genomic sequence consisted of two exons separated by a distance of 1.3 kb
